Biological Background: Megalin/LRP2 Function and Localization
- Megalin (LRP2) is a 521 kDa type I transmembrane glycoprotein belonging to the low-density lipoprotein receptor family, containing multiple ligand-binding repeats and EGF-like domains.
- Functions as a multiligand endocytic receptor, primarily at the apical surface of epithelial cells, mediating the uptake of lipoproteins, vitamins, hormones, and drugs in cooperation with cubilin.
- Mediates endocytosis and transport of leptin across the blood-brain barrier and within neuronal cells, contributing to hypothalamic leptin signaling and body weight regulation (By similarity).
- In the kidney, mediates renal reabsorption of 25-hydroxyvitamin D3 bound to GC/DBP, and clears metallothionein-bound heavy metals. Related references: PMID:15126248
- Plays a crucial role in kidney homeostasis by internalizing selenoprotein SEPP1 for selenium uptake and the antiapoptotic protein BIRC5/survivin for functional integrity. Related references: PMID:23825075
- Subcellular localization includes the apical cell membrane, endosome lumen, and coated pits; also present in neuronal dendrites and axons, indicating roles in polarized trafficking.
- Essential for embryonic development, including neural tube closure via folate receptor-mediated uptake and regulation of sonic hedgehog signaling.
- Expressed predominantly in absorptive epithelia such as renal proximal tubules and placental cytotrophoblasts, with implications for nutrient and steroid hormone transport.
Experimental Guidance and Technical Tips
- The immunogen maps to the C-terminal cytoplasmic domain (aa 4446-4655), making the antibody suitable for detecting intracellular epitopes after permeabilization.
- For immunofluorescence (IF-P), ensure adequate permeabilization (e.g., Triton X-100) to allow antibody access to the intracellular domain.
- For intracellular flow cytometry (FC intra), optimize fixation and permeabilization steps to maintain antigenicity; consider using a nuclear envelope marker for gating.
- The antibody cross-reacts with mouse Megalin, enabling studies in mouse models; however, perform initial validation in your specific tissue or cell type.
- Due to the protein’s large size (521 kDa), confirm specificity using knockdown (siRNA), knockout, or blocking peptide controls.
- Pair with anti-cubilin antibodies for co-localization studies, as both receptors are involved in endocytic processes in the proximal tubule.
