Biological Background: CD63 Function and Localization
- CD63 (also known as granulophysin, LAMP-3, ME491, OMA81H, Tspan-30) is a member of the tetraspanin family, a group of four-transmembrane domain proteins involved in cell adhesion, migration, and signal transduction.
- It functions as a receptor for tissue inhibitor of metalloproteinases 1 (TIMP1) and activates downstream signaling pathways including ITGB1/integrin signaling, leading to AKT, FAK/PTK2, and MAP kinase activation, promoting cell survival, adhesion, spreading, and migration.
- CD63 plays a role in VEGFA signaling by regulating internalization of KDR/VEGFR2, and is essential for normal PMEL luminal domain trafficking, critical for melanocyte development and maturation.
- It is implicated in leukocyte adhesion to endothelial cells through regulation of SELP trafficking, and may be involved in mast cell degranulation in response to specific stimuli.
- Subcellularly, CD63 is localized to the cell membrane, lysosome membrane, late endosome membrane, multivesicular bodies, melanosomes, and is also secreted in extracellular exosomes.
- Tissue specificity: detected in platelets, dysplastic nevi, radial growth phase primary melanomas, hematopoietic cells, and tissue macrophages.
- Keywords highlight its involvement in protein transport, alternative splicing, and post-translational modifications including glycosylation, palmitoylation, and lipidation.
Experimental Guidance and Technical Tips
- For flow cytometry, titrate the antibody to determine optimal staining concentration for your specific cell type and experimental conditions; a typical starting range is 0.1–1.0 µg per 10^6 cells.
- Include appropriate isotype controls and fluorescence-minus-one (FMO) controls to ensure specific staining, particularly for markers with heterogeneous expression like CD63.
- Since CD63 is expressed on the cell surface as well as intracellularly, consider permeabilization strategies if total or lysosomal staining is desired.
- Validate the antibody in your sample system, as cross-reactivity is confirmed for human only; other species may require additional testing.
