Biological Background: CD163 Function and Localization
- CD163, also known as M130 or scavenger receptor cysteine-rich type 1 protein, is a transmembrane protein expressed on macrophages.
- It serves as a receptor for hemoglobin/haptoglobin complexes, mediating their clearance and endocytosis to protect tissues from free hemoglobin-induced oxidative damage.
- Through endocytosis of hemoglobin/haptoglobin and subsequent heme breakdown, CD163 plays a role in iron recycling (By similarity).
- Ligand binding induces intracellular signaling involving tyrosine kinase-dependent calcium mobilization, inositol triphosphate production, and secretion of IL6 and CSF1.
- A soluble shed form (sCD163) may exert anti-inflammatory effects.
- CD163 is localized to the cell membrane and secreted, with tissue-specific expression in monocytes and mature macrophages such as Kupffer cells in the liver, red pulp macrophages in the spleen, and mesenteric lymph nodes.
- Structural features include scavenger receptor cysteine-rich domains, a transmembrane helix, disulfide bonds, and glycosylation; the protein is involved in acute phase and inflammatory responses and undergoes alternative splicing.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 39-260 in the extracellular domain, making this PE-conjugated antibody suitable for direct immunofluorescence detection in flow cytometry.
- Include an appropriate isotype control and, when working with mouse tissues, consider Fc receptor blocking to reduce non-specific binding.
- Titrate the antibody to determine the optimal staining concentration for your specific cell population.
- Validate antibody performance in a relevant mouse macrophage model, such as bone marrow-derived macrophages or tissue-resident macrophage preparations.
