Biological Background: LYAR Function and Localization
- LYAR (cell growth-regulating nucleolar protein, also known as ZLYAR or ZC2HC2) is a zinc-finger-containing protein with a predicted molecular weight of ~44 kDa; it functions as a DNA-binding transcriptional regulator.
- Plays a central role in pre-rRNA processing: maintains appropriate processing of 47S/45S pre-rRNA to 32S/30S pre-rRNAs and their subsequent conversion to mature 18S and 28S rRNAs (PubMed:24495227). Related references: PMID:24495227
- Acts as a transcriptional repressor: together with PRMT5, binds the gamma-globin (HBG1/HBG2) promoter and represses its expression; preferentially binds the DNA motif 5'-GGTTAT-3' (PubMed:25092918). Related references: PMID:25092918
- In neuroblastoma cells, represses the expression of oxidative stress genes, including CHAC1, HMOX1, SLC7A11, ULBP1 and SNORD41 (PubMed:28686580). Related references: PMID:28686580
- Negatively regulates the innate immune response by impairing IRF3 DNA-binding activity and inhibiting NF-kappa-B-mediated expression of pro-inflammatory cytokines (PubMed:31413131). Related references: PMID:31413131
- Localizes to the nucleus, nucleolus, cytoplasm, and cell projection cilium photoreceptor outer segment; predominantly expressed in testis.
- Additional functional roles: stimulates phagocytosis of photoreceptor outer segments by retinal pigment epithelial cells, and maintains nucleolin (NCL) protein stability in embryonic stem cells to support self-renewal (By similarity).
Experimental Guidance and Technical Tips
- For Western blotting, the predicted molecular weight of LYAR is 44 kDa; validate the observed band size in your sample system, as post-translational modifications or isoforms may cause shifts.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), use appropriate antigen retrieval and blocking steps; expect staining in nucleus/nucleolus and cytoplasm, reflecting LYAR localization patterns.
- For ELISA applications, the recombinant immunogen (amino acids 10-160) can be used as a positive control; consider knockout or knockdown validation to confirm antibody specificity.
- Since this is a rabbit polyclonal antibody, pair with suitable anti-rabbit secondary reagents; appropriate isotype controls are recommended for immunostaining.
