Biological Background: GARS Function and Localization
- GARS encodes glycyl-tRNA synthetase, a class II aminoacyl-tRNA synthetase that catalyzes the ATP-dependent attachment of glycine to its cognate tRNA during protein synthesis. Related references: PMID:17544401 PMID:24898252 PMID:28675565
- In addition to its role in translation, GARS functions as a diadenosine tetraphosphate synthetase (Ap4A synthetase), producing the signaling molecule Ap4A by condensation of two ATP molecules, thereby contributing to cellular regulation pathways. Related references: PMID:19710017
- The protein localizes to the cytoplasm, and is also found in mitochondria, cell projections (axons), and can be secreted via extracellular exosomes.
- GARS is widely expressed, with documented presence in brain, spinal cord, muscle, heart, spleen, and liver.
- Mutations in GARS are associated with Charcot-Marie-Tooth disease type 2D and distal spinal muscular atrophy type V, reflecting its importance in neuronal function.
- Post-translational modifications include acetylation and phosphorylation, and the protein contains an ATP-binding domain and a transit peptide for mitochondrial targeting.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide derived from the C-terminal region (amino acids 640–739) of human GARS; the antibody may recognize both full-length and truncated protein forms depending on sample context.
- For Western blot, an expected band around 83 kDa is anticipated in human samples; consider using appropriate positive controls and molecular weight markers.
- The antibody is validated for WB and ELISA; for ELISA, direct or sandwich formats using the immunogen peptide as standard may be explored.
- Cross-reactivity is reported for human; performance with other species has not been characterized and should be validated empirically if needed.
- Storage and handling: shipped on ice bags; upon receipt, aliquot and store at recommended temperatures to maintain stability.
