Biological Background: DAP Function and Localization
- Death-associated protein 1 (DAP/DAP1) is a small (11 kDa) protein encoded by the DAP gene in humans.
- Functions as a ribosome-binding protein involved in ribosome hibernation, stabilizing ribosomes in an inactive state and preventing proteasomal degradation (By similarity).
- Associates with eIF5a at the polypeptide exit tunnel of the ribosome to prevent mRNA translation, particularly in mature oocytes where ribosome inactivation is critical for early development (By similarity).
- Acts as a negative regulator of autophagy, a cellular degradation pathway. Related references: PMID:20537536
- Mediates interferon-gamma-induced cell death, linking to apoptotic pathways. Related references: PMID:7828849
- Post-translationally modified by acetylation and phosphorylation, as identified by proteomic studies.
Experimental Guidance and Technical Tips
- For Western blot, note the small predicted molecular weight of DAP1 (11 kDa); consider using a high-percentage gel and optimized transfer conditions to retain the protein.
- Validate antibody specificity in your sample system using positive and negative controls, especially when assaying tissues with low DAP expression.
- In immunohistochemistry, antigen retrieval may be necessary as the immunogen corresponds to a linear N-terminal epitope (amino acids 1-102).
- For immunocytochemistry, fixation and permeabilization conditions should be tailored to your cell type to ensure accessibility of the ribosomal target.
