Biological Background: BOB1 Function and Localization
- BOB1 is a transcriptional coactivator that specifically associates with POU2F1/OCT1 or POU2F2/OCT2, strongly enhancing POU2F1-mediated promoter activity and, to a lesser extent, POU2F2-mediated activity. Related references: PMID:7779176
- It recognizes the POU domains of POU2F1/OCT1 and POU2F2/OCT2, acting as a B-cell-specific coactivator. Related references: PMID:7859290
- BOB1 is essential for B-cell response to antigens and required for the formation of germinal centers. Related references: PMID:7623806, PMID:7859290
- It regulates IL6 expression in B cells as a coactivator of POU2F2/OCT2 (by similarity).
- BOB1 localizes to the nucleus, consistent with its role in transcription regulation.
- Tissue expression is B-cell specific; detected mainly in spleen, thymus, peripheral blood leukocyte, and small intestine. Related references: PMID:7779176, PMID:7859290
- Classified as an activator, proto-oncogene, and target of ubiquitin-like modification, BOB1 is implicated in chromosomal rearrangements and is identified by proteomics.
Experimental Guidance and Technical Tips
- The synthetic peptide immunogen spanning amino acids 150–250 has been designed to target a specific region of human BOB1; verify sequence identity in your species of interest if cross-reactivity is expected.
- For Western blot (WB), a band at approximately 27 kDa is predicted; consider using reducing conditions and a B-cell lysate (e.g., Raji or Daudi) as a positive control.
- ELISA applications can employ the immunogen peptide as a coating antigen or positive control to assess antibody binding.
- As a polyclonal antibody, lot-to-lot variability is possible; we recommend titrating the antibody and validating performance in your specific assay system.
- Additional applications may be feasible, but optimization and validation in the relevant sample matrix are advised.
