Biological Background: Bcl10 Function and Localization
- BCL10 functions as a critical adapter in immune signaling, bridging CARD domain-containing proteins (CARD9, CARD11, CARD14) to downstream activation of NF-kappa-B and MAP kinase p38 pathways, thereby regulating pro-inflammatory cytokine and chemokine expression. Related references: PMID:10187770, PMID:10364242, PMID:10400625
- Upon recruitment by homooligomerized CARD domain proteins, BCL10 polymerizes and forms the CBM complex with MALT1, leading to activation of NF-kappa-B and MAP kinase p38, essential for adaptive and innate immune responses. Related references: PMID:24074955, PMID:18287044, PMID:27777308
- BCL10 is activated by CARD9 downstream of C-type lectin receptors, playing a key role in antifungal immunity. Related references: PMID:26488816
- BCL10 is activated by CARD11 downstream of T-cell receptor (TCR) and B-cell receptor (BCR) signaling, linking antigen receptor activation to immune gene expression. Related references: PMID:18264101, PMID:18287044, PMID:24074955
- BCL10 promotes apoptosis and pro-caspase-9 maturation, and activates NF-kappa-B via NIK and IKK, indicating a role in cell death regulation. Related references: PMID:10187815
- The protein localizes to the cytoplasm, perinuclear region, and membrane rafts, consistent with its signaling roles at immune synapses.
- BCL10 is ubiquitously expressed and is subject to post-translational modifications including acetylation, phosphorylation, and ubiquitination (isopeptide bond).
- Clinically, BCL10 is associated with chromosomal rearrangements in lymphomas and has tumor suppressor properties, with disease variants linked to immune deficiencies.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-118) encompassing the CARD domain; this design may facilitate recognition of the native and recombinant protein, and can be suitable for detecting both full-length and potential splice variants.
- For Western blot (WB) detection, the expected molecular weight is approximately 26 kDa; consider using appropriate protein ladders and validate in the relevant sample system, as post-translational modifications may affect migration.
- For ELISA, titration of the antibody and inclusion of appropriate controls are recommended to optimize signal-to-noise ratios.
