Biological Background: ZNF683 Function and Localization
- ZNF683 (Hobit) is a zinc-finger transcription factor essential for the development and retention of tissue-resident memory T (Trm) cells, natural killer (NK) cells, and natural killer T (NKT) cells in non-lymphoid organs such as skin, gut, liver, and kidney. Related references: PMID:26179882
- It functions by binding to promoter regions of target genes, often in association with the transcriptional repressor PRDM1/BLIMP1, to negatively regulate genes that promote lymphocyte egress from tissues. Related references: PMID:26179882
- Isoform-specific activities have been reported: isoform 2 functions as a transcriptional repressor regulating interferon-gamma (IFN-γ) production, while isoform 1 lacks repressor activity but retains DNA-binding capability.
- ZNF683 is predominantly expressed in terminally differentiated effector CD8+ T-cells, NK cells, and NKT cells, with distinct isoform expression patterns. Related references: PMID:26179882, PMID:20921622
- During cytomegalovirus (CMV) infection, ZNF683 is upregulated in effector CD8+ T-cells, suggesting a role in antiviral immune responses. Related references: PMID:20921622
- Localized to the nucleus, ZNF683 belongs to the krueppel C2H2-type zinc-finger protein family and contains metal-binding zinc finger domains.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (aa 420-524) and may recognize multiple ZNF683 isoforms; validate specificity in your experimental system.
- For Western blot, the predicted molecular weight is approximately 57 kDa; verify band migration using appropriate positive controls.
- Consider testing the antibody on human lymphocyte subpopulations or cell lines known to express ZNF683.
- The antibody is suitable for WB and ELISA as indicated; optimize dilutions and conditions for other applications.
