Biological Background: USP9X/USP9Y Function and Localization
- USP9Y, also known as Ubiquitin carboxyl-terminal hydrolase 9Y, Deubiquitinating enzyme FAF-Y, or Fat facets protein-related Y-linked, is a member of the ubiquitin-specific protease (USP) family.
- Encoded by the USP9Y gene (alternative name DFFRY) on the Y chromosome, it functions as a deubiquitinase that cleaves ubiquitin from substrate proteins, playing a role in the ubiquitin-proteasome pathway. It is proposed to stabilize proteins important for spermatogenesis and male fertility. Related references: PMID:12895410
- Widely expressed in both embryonic and adult tissues.
- Key structural features include a zinc-binding domain typical of thiol proteases, and the protein is subject to phosphorylation and alternative splicing.
- The mature protein has a molecular weight of approximately 291 kDa.
- The antibody's immunogen sequence (amino acids 2456-2555) is shared between USP9Y and its X-chromosome homolog USP9X, enabling detection of both proteins.
Experimental Guidance and Technical Tips
- For Western blotting, expect a band around 291 kDa. Use a low-percentage polyacrylamide gel (e.g., 6–8%) and extended transfer times to ensure efficient transfer of high-molecular-weight proteins.
- Validate antibody specificity in your sample system by using siRNA knockdown or cell lines with known expression levels of USP9X/USP9Y.
- Consider peptide blocking experiments with the immunogen sequence to confirm band identity.
- For IHC-P, optimize antigen retrieval conditions (e.g., heat-induced epitope retrieval with citrate or EDTA buffer) and test different antibody dilutions.
- For IF/ICC, use appropriate fixation and permeabilization protocols for the subcellular localization of USP9X/USP9Y.
- As a polyclonal antibody, batch-to-batch consistency may vary; evaluate new lots against a reference standard.
