Biological Background: TTC7A Function and Localization
- TTC7A (Tetratricopeptide repeat protein 7A) is a 96 kDa protein encoded by the TTC7A gene (GeneID: 57217). It belongs to the TTC7 family and contains tetratricopeptide repeats (TPR) that mediate protein-protein interactions.
- TTC7A is a core component of a protein complex that localizes phosphatidylinositol 4-kinase (PI4K) to the plasma membrane. This complex regulates the synthesis of phosphatidylinositol 4-phosphate (PtdIns(4)P), a key lipid signaling molecule. Related references: PMID:23229899 PMID:24417819
- Within the complex, TTC7A acts as a central scaffold, bridging PI4KA to the membrane-anchoring proteins EFR3B and HYCC1 (By similarity).
- The protein localizes predominantly to the cytoplasm and cell membrane, consistent with its role in plasma membrane lipid signaling.
- Tissue-specific expression is observed in epithelial cells of the intestine, thymus, and pancreas, as confirmed at the protein level.
- TTC7A is subject to alternative splicing and phosphorylation, and mutations in the gene are associated with disease variants.
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 1-300 of human TTC7A (N-terminal region). This region may be conserved in rat, supporting observed cross-reactivity.
- For Western blot, a band at approximately 96 kDa is expected in human and rat cell/tissue lysates. Consider using positive controls like intestinal, thymic, or pancreatic epithelial cell lines.
- For ELISA, the antibody may be used as a capture or detection reagent; optimal dilutions should be determined empirically.
- Note that alternative splicing may generate isoforms; verify detection of the intended target in the relevant sample system.
- As a polyclonal antibody, it may recognize multiple epitopes; include appropriate negative controls to confirm specificity.
