Biological Background: STK3/MST2 Function and Localization
- Serine/threonine-protein kinase 3 (STK3), also known as mammalian STE20-like kinase 2 (MST2) or Krs-1, is a member of the STE20-like kinase family.
- Functions as a stress-activated, pro-apoptotic kinase; upon caspase cleavage, it localizes to the nucleus and triggers chromatin condensation and DNA fragmentation (PubMed:11278283, PubMed:8566796, PubMed:8816758). Related references: PMID:11278283 PMID:8566796 PMID:8816758
- Core component of the Hippo signaling pathway, where it phosphorylates and activates LATS1/2, leading to YAP1/TAZ inactivation and restricting cell proliferation and promoting apoptosis (PubMed:15688006, PubMed:16930133, PubMed:23972470). Related references: PMID:15688006 PMID:16930133 PMID:23972470
- Plays a role in centrosome disjunction by phosphorylating NEK2, thereby regulating its centrosomal localization and ability to phosphorylate CROCC and CEP250 (PubMed:21076410, PubMed:21723128). Related references: PMID:21076410 PMID:21723128
- Positively regulates RAF1 activation by suppressing its inhibitory phosphorylation on Ser-259 (PubMed:20212043). Related references: PMID:20212043
- Phosphorylates MOBKL1A, MOBKL1B, and RASSF2, and cooperates with MOBKL1B to activate STK38 (PubMed:18328708, PubMed:18362890, PubMed:19525978). Related references: PMID:18328708 PMID:18362890 PMID:19525978
- Localizes to the cytoplasm, nucleus, and centrosome; its subcellular distribution is critical for its diverse functions.
- Expressed at high levels in adult kidney, skeletal muscle, and placenta, and at very low levels in heart, lung, and brain.
- Listed keywords include acetylation, alternative splicing, and Ubl conjugation, indicating post-translational modifications that may regulate its activity.
Experimental Guidance and Technical Tips
- For Western blotting, the predicted molecular weight is approximately 56 kDa; however, post-translational modifications may cause bands at different sizes. Include appropriate positive and negative controls.
- In immunofluorescence/ICC, since STK3 localizes to cytoplasm, nucleus, and centrosome, ensure adequate permeabilization and consider co-staining with organelle markers to confirm localization.
- For ELISA, the antibody is suitable; optimize coating and detection conditions based on the assay format.
- The immunogen corresponds to amino acids 323-450, covering the C-terminal region, which may recognize both full-length and caspase-cleaved fragments. Validate in relevant sample systems (e.g., HeLa, HEK293, or kidney-derived cell lines).
