Biological Background: RTN3 Function and Localization
- RTN3, also known as HAP, ASYIP, NSPL2, NSPLII, or RTN3-A1, is a member of the reticulon family with a predicted molecular weight of approximately 113 kDa (112611 Da).
- It primarily localizes to the endoplasmic reticulum and Golgi apparatus membranes, where it regulates membrane trafficking in the early secretory pathway.
- RTN3 inhibits BACE1 activity and amyloid precursor protein (APP) processing, a pathway implicated in Alzheimer's disease.
- RTN3 promotes the formation of endoplasmic reticulum tubules. Related references: PMID:25612671
- It plays a role in apoptosis by inducing the caspase-8 cascade and favoring BCL2 translocation to mitochondria under ER stress.
- As an inflammation-resolving regulator, RTN3 interacts with TRIM25 and RIGI, impairing RIGI ubiquitination and inhibiting IRF3 and NF-kappa-B signaling.
- Isoform 1 is highly expressed in brain and low in skeletal muscle; isoform 2 is restricted to melanoma; isoform 3 is widely expressed, with highest levels in brain neuronal cell bodies.
- RTN3 is also involved in host-virus interactions, playing a positive role in the replication and pathogenesis of enteroviruses.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to residues 100 to the C-terminus of human RTN3 (NP_006045.1); the antibody may recognize multiple isoforms sharing this region.
- For Western blot, expect a specific band around 113 kDa in human tissue or cell lysates. Include appropriate positive and negative controls.
- In ELISA, consider coating with the immunizing peptide or recombinant protein and titrating the antibody to optimize signal.
- Validate antibody specificity in relevant samples, such as human brain lysates, where RTN3 is highly expressed.
