Biological Background: PARL Function and Localization
- PARL (Presenilin-associated rhomboid-like protein, mitochondrial) is a mitochondrial intramembrane cleaving protease, also known as PSARL, RHBDS1, or PRO2207.
- It is required for apoptosis control during postnatal growth, essential for proteolytic processing of anti-apoptotic OPA1, which prevents mitochondrial cytochrome c release in response to intrinsic apoptotic signals (by similarity).
- Mediates maturation of PINK1 into its 52kDa form after cleavage by MPP (PubMed:22354088). Related references: PMID:22354088
- Promotes cleavage of PGAM5 in damaged mitochondria in response to mitochondrial membrane potential loss (PubMed:22915595). Also mediates differential cleavage of PINK1 and PGAM5 depending on mitochondrial health status (PubMed:22915595). Related references: PMID:22915595
- Required for processing CLPB into a form with higher protein disaggregase activity by removing an autoinhibitory N-terminal peptide (PubMed:28288130, PubMed:32573439). Related references: PMID:28288130 PMID:32573439
- Promotes processing of DIABLO/SMAC in the mitochondrion required for its apoptotic activity, and mediates STARD7 processing to regulate its mitochondrial-cytosolic localization (PubMed:28288130, PubMed:29301859, PubMed:36658222). Also cleaves TTC19 (PubMed:28288130). Related references: PMID:28288130 PMID:29301859 PMID:36658222
- Promotes changes in mitochondria morphology regulated by phosphorylation of P-beta domain (PubMed:14732705, PubMed:17116872). Related references: PMID:14732705 PMID:17116872
- Localizes to the mitochondrial inner membrane and can also be found in the nucleus.
- Involved in alternative splicing; classified as a serine protease with a transit peptide and multiple transmembrane helices.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-170) of human PARL, which may target the full-length protein and potentially processed forms.
- For WB, consider that PARL has a predicted molecular weight of 42 kDa; however, due to cleavage and processing, multiple bands may be observed. Validate the expected band size in your sample system.
- Because PARL is a membrane protein, use appropriate lysis buffers and consider sample preparation conditions for mitochondrial enrichment if needed.
- For IF/ICC, confirm localization to mitochondrial structures; co-staining with mitochondrial markers is recommended.
- ELISA applications may require optimization of coating conditions; consider using recombinant protein as standard.
- Cross-reactivity is indicated for human; for other species, check sequence homology before use.
