Biological Background: NUP98 Function and Localization
- NUP98, also known as ADIR2, NUP96, NUP196, or Nup98-96, is a component of the nuclear pore complex (NPC) and is produced as a precursor protein that undergoes autocatalytic cleavage to generate NUP98 and NUP96, both integral to NPC function.
- It plays a critical role in NPC assembly and maintenance, and both NUP98 and NUP96 are required for bidirectional nucleocytoplasmic transport. Related references: PMID:33097660
- In cooperation with DHX9, NUP98 participates in transcription regulation and alternative splicing of specific genes, and is involved in localizing DHX9 to discrete intranuclear foci known as GLFG bodies. Related references: PMID:28221134
- The protein localizes to the nuclear membrane and is predominantly found at the nuclear pore complex, with additional presence in the nucleoplasm.
- Key post-translational modifications include acetylation, phosphorylation, isopeptide bond formation, and ubiquitin-like conjugation, which may regulate its function and interactions.
- NUP98 interacts with HIV-1 capsid protein P24 and nucleocapsid protein P7, potentially facilitating viral integration into the host nucleus, with reduced binding affinity observed for certain capsid mutations. Related references: PMID:23523133
- The protein features FG/GLFG repeat domains that serve as docking sites for transport receptors, enabling selective passage of macromolecules through the NPC.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a region within the NUP98 portion (aa 621-920) of the full-length NUP98 precursor, suggesting the antibody primarily detects the cleaved NUP98 form, but may also recognize the precursor due to sequence overlap.
- For Western blotting, a band around 98 kDa (cleaved NUP98) or 198 kDa (precursor) may be observed depending on sample preparation and electrophoresis conditions; validate using nuclear extracts from human or mouse cells.
- For immunofluorescence/ICC, expect punctate nuclear rim staining characteristic of nuclear pore complexes; permeabilization is necessary for antibody access.
- ELISA applications can be used for quantifying NUP98 levels in cell lysates or purified fractions; optimize coating and detection conditions empirically.
