Biological Background: MRGPRX1 Function and Localization
- MRGPRX1 (Mas-related G protein-coupled receptor member X1), also known as sensory neuron-specific G protein-coupled receptor 3/4, is a member of the Mas-related GPCR family and functions as a receptor for various pruritogens and pain modulators.
- It is critically involved in itch perception and pain transmission, specifically in sensory neurons of the dorsal root ganglion and trigeminal ganglia (PubMed:11850634, PubMed:20004959, PubMed:26582731). Related references: PMID:11850634 PMID:20004959 PMID:26582731
- MRGPRX1 is activated by diverse ligands: enkephalin neuropeptides like BAM22 and BAM8-22 (PubMed:11850634, PubMed:36302898, PubMed:37591889), tick defensins (PubMed:33358893), conotoxin CNF-Tx2 (PubMed:30243794, PubMed:37591889), chloroquine (PubMed:20004959), and dust mite cysteine protease Der p1 (PubMed:28768771). Related references: PMID:11850634 PMID:36302898 PMID:37591889 PMID:33358893 PMID:30243794 PMID:20004959 PMID:28768771
- Ligand binding triggers signaling via dual coupling to G(q) and G(i) proteins, leading to phospholipase C-beta activation and adenylate cyclase inhibition, respectively, which modulates downstream effectors including TRPV1 sensitization (PubMed:23074220, PubMed:36302898, PubMed:37347749). Related references: PMID:23074220 PMID:36302898 PMID:37347749
- Activation in the peripheral nervous system elicits scratching behavior, while in the central nervous system it dampens chronic pain by attenuating spinal nociceptive transmission (PubMed:28223516). It also mediates sneezing responses in the nasal mucosa upon activation by irritants or influenza virus. Related references: PMID:28223516
- MRGPRX1 is localized to the cell membrane and is specifically expressed in small-diameter sensory neurons of the dorsal root and trigeminal ganglia.
- Key structural features include seven-transmembrane helices, disulfide bonds, glycosylation sites, and a conserved GPCR fold, as inferred from its classification as a G protein-coupled receptor (UniProt keywords: 3D-structure, Acute phase, Cell membrane, Disulfide bond, G protein-coupled receptor, Glycoprotein, Membrane, Receptor, Transducer, Transmembrane, Transmembrane helix).
Experimental Guidance and Technical Tips
- For Western blotting, expect a band at approximately 36 kDa corresponding to the predicted molecular weight of MRGPRX1. Consider optimizing reducing conditions and validating in dorsal root ganglion lysates or transfected cell lines.
- The immunogen corresponds to the C-terminal region (aa 263-322), which may influence epitope accessibility; pre-adsorption with the immunogen peptide can serve as a specificity control.
- For ELISA, the recombinant protein immunogen provides good reactivity; ensure coating conditions are optimized and validate with known positive and negative samples.
- Given cross-reactivity with mouse and rat, this antibody can be used in rodent pain and itch models, but confirm epitope conservation across species.
