Biological Background: HADHB Function and Localization
- Trifunctional enzyme subunit beta, mitochondrial (also known as TP-beta), is encoded by the HADHB gene in humans.
- It is part of the mitochondrial trifunctional enzyme complex, a heterotetramer with HADHA, catalyzing the last three steps of mitochondrial long-chain fatty acid beta-oxidation: hydration, dehydrogenation, and thiolysis. Related references: PMID:1550553, PMID:16423905, PMID:26474213
- HADHB specifically catalyzes the 3-ketoacyl-CoA thiolase activity, cleaving 3-ketoacyl-CoA into acetyl-CoA and a shortened acyl-CoA, thereby completing the beta-oxidation cycle. Related references: PMID:16423905, PMID:26474213, PMID:29915090
- The enzyme has been experimentally confirmed to act on saturated long-chain fatty acids such as palmitate (hexadecanoate) and laurate (dodecanoate), and is predicted to also process unsaturated fatty acids like oleate and linoleate. Related references: PMID:1550553, PMID:8135828, PMID:8163672, PMID:26474213
- Subcellular localization includes the mitochondrial inner membrane, mitochondrial outer membrane, and endoplasmic reticulum, reflecting its role in lipid metabolism.
- The protein is subject to post-translational modifications including acetylation, and alternative splicing produces multiple isoforms; mutations in HADHB are associated with mitochondrial trifunctional protein deficiency (a disease phenotype).
- Each beta-oxidation cycle yields acetyl-CoA, FADH₂, and NADH, contributing to ATP production in energy-demanding tissues. Related references: PMID:26474213, PMID:29915090
Experimental Guidance and Technical Tips
- The immunogen used is a recombinant fusion protein comprising amino acids 34-270 of human HADHB. For Western blot (WB), a band of approximately 51 kDa is expected; validate the antibody in your specific sample system and optimize dilution.
- For Immunofluorescence (IF/ICC), the mitochondrial localization of HADHB should be observable; co-staining with mitochondrial markers is recommended to confirm specificity.
- The antibody shows cross-reactivity with human, mouse, and rat, making it suitable for studies in these species; confirm sequence homology if other species are to be tested.
- ELISA applications may benefit from checkerboard titration to determine optimal coating and detection conditions.
- As a polyclonal antibody, lot-to-lot variability may occur; include positive and negative controls in each experiment.
