Biological Background: G2E3 Function and Localization
- G2E3, also known as PHF7B or KIAA1333, is a G2/M phase-specific HECT-type E3 ubiquitin-protein ligase.
- It catalyzes the monoubiquitination of histone H3 at Lys-14 (H3K14ub), a mark essential for heterochromatin formation at pericentric regions. Related references: PMID:41094145
- H3K14ub facilitates the deposition of H3K9me3 and subsequent recruitment of SUV39H and HP1 proteins, thereby establishing constitutive heterochromatin. Related references: PMID:41094145
- G2E3 associates with mitotic chromosomes in an RNA-dependent manner to perform its ubiquitination function.
- Subcellularly, it localizes to the nucleus, chromosomes, and nucleolus, with some cytoplasmic presence.
