Biological Background: FMRP Function and Localization
- FMRP is a multifunctional polyribosome-associated RNA-binding protein essential for neuronal development and synaptic plasticity. It regulates alternative mRNA splicing, mRNA stability, dendritic mRNA transport, and postsynaptic local protein synthesis. Related references: PMID:12417522, PMID:16631377, PMID:18653529
- Acts as a repressor of mRNA translation, particularly at synapses, by forming neuronal ribonucleoprotein granules and recruiting EIF4EBP2; it undergoes liquid-liquid phase separation upon target mRNA binding and phosphorylation. Related references: PMID:30765518, PMID:31439799
- Binds to G-quadruplex structures and specific RNA motifs such as 5'-ACU[GU]-3' and 5'-[AU]GGA-3' in target mRNAs, mediating translation regulation and mRNA localization. Related references: PMID:11719189, PMID:23235829, PMID:25464849
- Localizes to multiple subcellular compartments: cytoplasm, stress granules, neuronal projections (axons, dendrites, dendritic spines), synapses, nucleus, and Cajal bodies, reflecting its roles in mRNA transport, translation, and nuclear export.
- Implicated in mRNA nuclear export by specifically recognizing N6-methyladenosine (m6A)-containing mRNAs and promoting their export in a CRM1-dependent manner. Related references: PMID:31753916
- Highly expressed in brain, cerebellum, testis, and also found in epithelial tissues, neurons, Purkinje cells, spermatogonia, oligodendrocytes, and lymphocytes. Related references: PMID:8401578, PMID:9259278, PMID:23891804, PMID:8504300
- Post-translational modifications including phosphorylation, acetylation, methylation, and ubiquitination regulate FMRP activity, phase separation, and protein interactions.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 500–600 of human FMRP; the antibody may recognize full-length FMRP and any isoforms containing this region. Validate specificity in your experimental system.
- For western blot (WB), a band near 71 kDa is expected, though post-translational modifications or isoform variations may shift migration. Include appropriate controls.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), consider using tissue or cell types with high FMRP expression (e.g., brain, testis) and optimize fixation/permeabilization conditions.
- For ELISA, peptide-based detection may be optimized; consider pairing with a validated capture/detection system.
- Cross-reactivity is reported for human, mouse, and rat. Validate in the relevant species, as sequence homology in the immunogen region may vary.
