Biological Background: EB3/MAPRE3 Function and Localization
- EB3/MAPRE3 (Microtubule-associated protein RP/EB family member 3), also known as EB1 protein family member 3 (EBF3), End-binding protein 3 (EB3), or RP3, is encoded by the MAPRE3 gene and belongs to the RP/EB family of microtubule plus-end tracking proteins (+TIPs).
- It functions as a +TIP that binds to the plus-ends of microtubules and promotes microtubule growth, thereby regulating the dynamics of the microtubule cytoskeleton (PubMed:19255245, PubMed:28814570). Related references: PMID:19255245 PMID:28814570
- EB3/MAPRE3 is involved in spindle function by stabilizing microtubules and anchoring them at centrosomes, contributing to proper cell division (PubMed:19255245, PubMed:28814570). Related references: PMID:19255245 PMID:28814570
- Additionally, it acts as a regulator of minus-end microtubule organization by interacting with the AKAP9-PDE4DIP complex and recruiting CAMSAP2 to the Golgi apparatus, tethering non-centrosomal minus-end microtubules to the Golgi for polarized cell movement (PubMed:28814570). Related references: PMID:28814570
- EB3/MAPRE3 localizes to the cytoplasm and associates with the cytoskeleton.
- It is predominantly expressed in brain and muscle tissues.

Experimental Guidance and Technical Tips
- This antibody was generated against a synthetic peptide corresponding to amino acids 182-281 of human EB3/MAPRE3 (Q9UPY8), targeting a region near the C-terminus. When designing experiments, verify that the immunogen sequence is conserved in your species of interest.
- For Western blot (WB), the predicted molecular weight of EB3/MAPRE3 is approximately 32 kDa (UniProt: 31.98 kDa). A band near this size is expected; always include appropriate positive and negative controls.
- In immunofluorescence/immunocytochemistry (IF/ICC), EB3/MAPRE3 localizes to microtubule plus-ends. Consider using fixation methods that preserve cytoskeletal structures (e.g., methanol or paraformaldehyde with detergent) and co-staining with tubulin markers to confirm specificity.
- This antibody is indicated for ELISA applications. Optimize coating antigen and antibody dilution for your specific assay format.
- The antibody cross-reacts with human, mouse, and rat species. Validate performance in the relevant model system, as epitope sequence variations may affect binding.
