Biological Background: DDX17 Function and Localization
- Encodes the DEAD box RNA helicase p72 (probable ATP-dependent RNA helicase DDX17), also known as p82, involved in diverse aspects of RNA metabolism and gene regulation.
- Functions as an ATP-dependent RNA helicase that unwinds RNA and modulates RNA structures, playing a pivotal role in pre-mRNA splicing and alternative splicing of exons with specific features, such as AC-rich exons in CD44 transcripts.

Related references: PMID:24581491, PMID:27478153, PMID:25126784, PMID:17485482 - Acts as a context-dependent transcriptional coregulator; enhances NFAT5 activity, synergizes with TP53 to activate MDM2 (requires acetylation), coactivates MMP7, and with CTNNB1 coactivates MYC, JUN, FOSL1, and CCND1 transcription.
Related references: PMID:15298701, PMID:22266867, PMID:17226766, PMID:19995069, PMID:20663877, PMID:17699760 - Plays critical roles in steroid hormone signaling; regulates alternative promoter usage and splicing of estrogen-responsive genes, and through helicase-independent mechanisms, coactivates estrogen receptor ESR1.
Related references: PMID:19995069, PMID:20406972, PMID:20663877, PMID:19718048, PMID:24275493 - Contributes to innate antiviral immunity by restricting bunyavirus infection (RVFV, LACV) in an interferon-independent manner; binds viral RNA and promotes ZC3HAV1-mediated mRNA degradation in an ATPase-dependent mechanism.
Related references: PMID:25126784, PMID:18334637 - Localizes to the nucleus and nucleolus, with additional cytosolic presence. Widely expressed, but protein levels increase during colon cancer progression from benign polyps to adenomas.
- Underwent multiple post-translational modifications including acetylation, methylation, phosphorylation, and ISGylation; also subject to alternative initiation and splicing, contributing to functional diversity.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 100–200 within the N-terminal region of human DDX17. This region is conserved among human, mouse, and rat, supporting the observed cross-reactivity. Consider mapping the epitope for precise binding characterization.
- For western blot (WB), a band of approximately 80 kDa is expected. Use appropriate positive controls (e.g., HeLa, HEK293) and verify specificity by siRNA knockdown if needed.
- In immunohistochemistry (IHC-P), nuclear staining is anticipated; validation in colon cancer progression tissues may reveal differential expression as reported in literature. Optimize antigen retrieval and antibody dilution.
- For ELISA applications, the antibody may be used as a capture or detection reagent; pair with a matched DDX17 detection antibody for sandwich ELISA. Validate performance with recombinant DDX17 protein.
- This monoclonal antibody may be suitable for immunoprecipitation (IP) or chromatin immunoprecipitation (ChIP) given the target’s roles in transcription and RNA binding, though not listed in applications; pilot testing is recommended.
- Monitor shipping conditions upon receipt; aliquoting and storage at -20°C is recommended for long-term stability.
