Biological Background: Corticotropin-Releasing Hormone (CRH) Function and Localization
- CRH, also known as corticotropin-releasing factor (CRF) or corticoliberin, is a secreted peptide hormone primarily produced by the hypothalamus and placenta.
- It plays a central role in whole-body adaptation to stress by binding to CRHR1 and, to a lesser extent, CRHR2. This triggers the neuroendocrine, autonomic, and behavioral stress responses, including activation of the hypothalamic-pituitary-adrenal (HPA) axis and subsequent adrenocorticotropic hormone (ACTH) release. Related references: PMID:18801728 PMID:3003585 PMID:32004469
- CRH acts as a "placental clock" during pregnancy, regulating gestational length, fetal development, and the timing of labor. Related references: PMID:7585095
- It also regulates blastocyst implantation by promoting decidualization, regulating trophoblast invasion, and facilitating maternal immune tolerance. Related references: PMID:11590404
- The CRH prohormone undergoes post-translational modifications including amidation and cleavage on pair of basic residues, which are critical for generating the mature, biologically active hormone.
- As a secreted protein, CRH is released into the extracellular space and is detectable in blood and cerebrospinal fluid.
- Beyond its endocrine functions, CRH is implicated in immune modulation, appetite regulation, and behavioral processes.
Experimental Guidance and Technical Tips
- The immunogen, spanning amino acids 25-194 of the mature CRH peptide, is designed to generate antibodies against the biologically active hormone. Consider verifying specificity using a blocking peptide if needed.
- For Western blot, the predicted molecular weight is approximately 21 kDa; a band near this size in hypothalamus or placenta lysates can be expected. Inclusion of protease inhibitors is recommended due to rapid hormone processing.
- In ELISA, titrate the antibody under your specific assay conditions and validate against known CRH standards. Cross-reactivity with mouse and rat CRH allows use in rodent stress models.
- Given the secretory nature of CRH, sample enrichment or concentration may improve detection in low-abundance fluids like serum or culture supernatants.
