Biological Background: CPA6 Function and Localization
- Carboxypeptidase A6 (CPA6, also known as CPAH, ETL5, FEB11) is a secreted, zinc-dependent metallocarboxypeptidase that belongs to the peptidase M14 family.
- It is implicated in the proteolytic processing of neuropeptides; specifically, it may inactivate enkephalins and neurotensin in certain brain regions and can convert angiotensin I to the active angiotensin II. Related references: PMID:18178555
- CPA6 preferentially cleaves C-terminal large hydrophobic amino acids, exhibiting minimal activity toward small residues or histidine. Related references: PMID:20855895
- The gene undergoes alternative splicing and chromosomal rearrangements have been observed; mutations in CPA6 are associated with familial febrile seizures and epilepsy, as indicated by its alias FEB11.
- Subcellularly, CPA6 is directed to the extracellular space and extracellular matrix, consistent with its role as a secreted enzyme.
- Expression is predominantly detected in the hippocampus, nucleus raphe, and cortex, supporting its involvement in neurological functions.
- Post-translational modifications include signal peptide cleavage, zymogen activation by removal of a pro-domain, N-glycosylation, and disulfide bond formation.
Experimental Guidance and Technical Tips
- The immunogen (amino acids 130–437) corresponds to the catalytic domain of human CPA6, suggesting that the antibody may recognize both pro- and active forms, as well as truncated variants; verify performance in relevant sample systems.
- For Western blot (WB), a band around 51 kDa is expected; consider using reducing and non-reducing conditions to assess potential disulfide-mediated dimerization or aggregation.
- In IHC-P, paraffin-embedded tissue sections may require antigen retrieval; optimal staining has been observed in brain tissues (hippocampus, cortex), but validate with appropriate controls.
- This polyclonal antibody may exhibit batch-to-batch variability; it is advisable to titrate the antibody and include blocking with the immunogen or a specific peptide for competition assays.
- For ELISA, the antibody can be used as a capture or detection reagent; cross-reactivity with human, mouse, and rat CPA6 supports multi-species assay development.
