Biological Background: CDKN2C/p18-INK4C Function and Localization
- CDKN2C encodes cyclin-dependent kinase 4 inhibitor C, also known as p18-INK4c or p18-INK6, a member of the INK4 family of CDK inhibitors.
- The protein contains ankyrin repeats (ANK repeat), a motif involved in protein-protein interactions.
- p18-INK4C strongly interacts with CDK6 and weakly with CDK4, inhibiting kinase activity to regulate cell cycle progression.
- Inhibition of CDK4/6 by p18-INK4C suppresses phosphorylation of the retinoblastoma protein (RB), thereby blocking cell cycle entry from G1 to S phase and inhibiting cell proliferation.
- Highest expression of CDKN2C is observed in skeletal muscle, with lower levels in pancreas and heart, indicating tissue-specific roles in cell cycle control.
- The protein has a molecular weight of approximately 18 kDa, consistent with its designation as p18.
Experimental Guidance and Technical Tips
- The immunogen encompasses the full-length human CDKN2C protein; the antibody is expected to recognize both native and denatured forms.
- For Western blotting, a band around 18 kDa is anticipated; consider using cell lines with high CDKN2C expression, such as those derived from skeletal muscle or pancreatic tissues, as positive controls.
- In ELISA, the antibody may be applied for detection of recombinant or purified CDKN2C; appropriate secondary detection systems should be validated.
- Cross-reactivity with mouse is reported; verify performance in your specific experimental system.
