Biological Background: Cas9 Function and Localization
- Cas9 (CRISPR-associated endonuclease Cas9/Csn1) is an RNA-guided DNA endonuclease that serves as the effector protein of the type II CRISPR-Cas adaptive immune system in Streptococcus pyogenes.
Related references: PMID:21455174 - In the CRISPR interference pathway, Cas9 forms a complex with a CRISPR RNA (crRNA) and a trans-activating crRNA (tracrRNA). This dual-RNA guided complex recognizes and cleaves double-stranded DNA targets complementary to the crRNA spacer sequence.
- Target recognition and cleavage require a short protospacer adjacent motif (PAM) sequence (5’-NGG-3’ for SpCas9) immediately downstream of the target site. PAM recognition is essential for catalytic activity and helps distinguish self from non-self DNA.
Related references: PMID:24476820 - Beyond DNA cleavage, Cas9 also participates in the maturation of crRNAs. Together with tracrRNA and host ribonuclease III, it stabilizes the pre-crRNA:tracrRNA duplex for correct processing, though Cas9 itself has no catalytic role in RNA cleavage.
Related references: PMID:24270795 - Cas9 is a multi-domain protein with DNA-binding, endonuclease, and exonuclease activities. It is dependent on divalent metal ions such as magnesium or manganese for its nucleolytic functions.
- Structurally characterized at high resolution, Cas9 exhibits conformational flexibility that enables its roles in RNA binding, DNA unwinding, and coordinated cleavage by its two nuclease domains (RuvC and HNH).
- The native molecular weight of Cas9 is approximately 158 kDa, consistent with its large multi-domain architecture.
Experimental Guidance and Technical Tips
- The immunogen comprises amino acids 450–730 of Streptococcus pyogenes Cas9, a region that includes portions of the RuvC nuclease domain. Consider that the antibody may recognize both native and denatured Cas9 protein.
- For Western blotting, the predicted molecular weight of full-length Cas9 is ~158 kDa; validate reactivity in the relevant sample system (e.g., S. pyogenes lysates or engineered cell lines expressing SpCas9).
- When developing an ELISA, evaluate the antibody’s performance against recombinant Cas9 protein or cell extracts. The indicated cross-reactivity (Human, Other) suggests potential for applications involving human cell lines or tissues expressing exogenous Cas9.
