Biological Background: BMPR2 Function and Localization
- BMPR2 (Bone morphogenetic protein receptor type-2, BMPR-II) is a transmembrane serine/threonine kinase receptor, with a molecular weight of ~115 kDa, and belongs to the transforming growth factor beta (TGF-β) receptor superfamily.
- Upon ligand binding (e.g., BMP2, BMP7), BMPR2 forms a heterotetrameric complex with type I receptors, leading to phosphorylation and activation of the type I receptor, which subsequently phosphorylates SMAD transcription factors (SMAD1/5/8) to regulate gene expression.
- BMPR2 can also mediate non-canonical signaling through the p38MAPK cascade, as demonstrated in certain cellular contexts.
Related references: PMID:12045205 - BMPR2 binds BMP7 and BMP2 with high affinity, BMP4 less efficiently, and this binding is enhanced by the presence of type I receptors. Additionally, it can signal via activin A/INHBA.
Related references: PMID:24018044 - The receptor is localized primarily at the cell membrane.
- It is highly expressed in heart and liver tissues.
- BMPR2 is subject to post-translational modifications including glycosylation and phosphorylation, as indicated by keywords: Glycoprotein, Phosphoprotein.
- Mutations in BMPR2 are associated with pulmonary arterial hypertension (PAH), reflecting its role as a disease variant biomarker.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (aa 27-150) of human BMPR2, suggesting the antibody likely recognizes the extracellular region. This may be suitable for detecting both full-length and soluble receptor forms.
- For Western blot (WB), the expected band size is ~115 kDa for the full-length receptor. Consider using appropriate molecular weight markers and validate with relevant positive control cell lysates (e.g., heart or liver tissue).
- In immunofluorescence (IF-P) and immunohistochemistry (IHC-P), BMPR2 is localized to the cell membrane; ensure proper permeabilization and include membrane marker co-staining for specificity validation.
- Cross-reactivity is confirmed with human, mouse, and rat species. For experiments using other species, sequence homology alignment of the immunogen region is recommended.
- For ELISA applications, the antibody can be used as a capture or detection reagent; optimize coating concentrations and validate with recombinant BMPR2 protein standards.
- Since BMPR2 signaling involves SMAD and p38MAPK pathways, functional studies may benefit from combining this antibody with phospho-specific antibodies for downstream signaling analysis.
