Biological Background: AXL Function and Localization
- AXL (UFO, Tyro7) is a receptor tyrosine kinase that binds growth arrest-specific protein 6 (GAS6), regulating cell survival, proliferation, migration, differentiation, and innate immune suppression.
- Ligand-induced dimerization leads to autophosphorylation and activation of downstream pathways including PI3K/AKT, GRB2, and PLCG1, which are crucial for endothelial survival, hepatic regeneration, and platelet activation.
- AXL inhibits Toll-like receptor (TLR)-mediated innate immune responses and contributes to natural killer cell development.
- As a viral entry receptor, AXL facilitates infection by Lassa, lymphocytic choriomeningitis, Ebola, and Zika viruses through GAS6-dependent recognition of phosphatidylserine on viral envelopes. For Zika, AXL also suppresses type I interferon signaling via SOCS1. Related references: PMID:28076778 PMID:29379210 PMID:31311882
- AXL is localized at the cell membrane and is highly expressed in metastatic colon tumors, with weaker expression in normal colon tissue, highlighting its role as an oncogene.
- Alternative splicing and post-translational modifications (glycosylation, phosphorylation, ubiquitination) further regulate AXL function.
- The protein has a molecular weight of approximately 98 kDa (UniProt: 98337 Da).
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular region (aa 33–449), which is expected to recognize both full-length and soluble forms of AXL. Consider this when designing assays for soluble AXL detection.
- For WB, AXL migrates at ~98 kDa; ensure adequate separation and transfer for high molecular weight proteins.
- For IHC-P, antigen retrieval and permeabilization may be needed; validate in tissues known to express AXL (e.g., metastatic colon cancer samples).
- For viral entry studies, this antibody may be used to detect AXL expression in permissive cell lines.
- Since it is a polyclonal, it may recognize multiple epitopes; validate specificity using knockdown or knockout controls.
- Use in ELISA and IP has been demonstrated under standard conditions; titrate antibody concentration for optimal signal.
