Biological Background: ACADM Function and Localization
- ACADM (medium-chain specific acyl-CoA dehydrogenase, also known as MCAD, ACAD1, MCADH) catalyzes the stereospecific alpha,beta-dehydrogenation of fatty acyl-CoA thioesters using electron transfer flavoprotein (ETF) as acceptor, preferring saturated C6–C12 chains and longer chains up to C16. Related references: PMID:1970566 PMID:21237683 PMID:2251268
- Electrons from reduced ETF are transferred to the mitochondrial respiratory chain via ETF-ubiquinone oxidoreductase, linking fatty acid oxidation to ATP production. Related references: PMID:15159392 PMID:25416781
- ACADM localizes to the mitochondrial matrix, consistent with its role in the beta-oxidation pathway.
- Exhibits ubiquitous expression, with highest levels detected in heart and skeletal muscle tissues.
- The enzyme is a flavoprotein (FAD-containing) and undergoes regulatory post-translational modifications including acetylation and phosphorylation.
- Mutations in the ACADM gene are linked to medium-chain acyl-CoA dehydrogenase (MCAD) deficiency, an inherited metabolic disorder.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide covering amino acids 200–300 of human ACADM; this rabbit monoclonal antibody offers reproducible affinity and specificity.
- In western blot (WB), a major band near 47 kDa is expected; observe for any additional bands that might represent processing intermediates or modified forms.
- For immunohistochemistry (paraffin, IHC-P) and immunofluorescence (IF/ICC), consider mitochondrial-targeted antigen retrieval and permeabilization protocols.
- ELISA applications should be optimized with matched antibody pairs and validated recombinant ACADM protein standards.
- Reported cross-reactivity covers human, mouse, and rat samples; confirm epitope conservation before applying to other species.
